This emphatic win positioned Japan at the summit of Group C on six points, after their earlier 2-0 success against Taiwan in the opening match.
They have netted 13 goals without conceding any.
Taiwan and Vietnam sit on three points apiece, leaving India stranded at the bottom with zero.
Taiwan boosted their chances of advancing by securing a narrow 1-0 triumph over Vietnam amid scorching conditions in Perth earlier that day.
Su Yu-hsuan delivered the decisive strike in the 26th minute, heading home from near point-blank range.
Having started with a 2-1 win over India, Vietnam faced a tougher challenge from Taiwan in heat nearing 40C, before a small audience under intense sun.
Vietnam increased their efforts late on but managed only one effort on target throughout.
North Korea, South Korea, hosts Australia, and defending champions China have all qualified for the quarter-finals with one match remaining.
